Data Mining On Machine Breakdowns And Effectiveness Of Scheduled Maintenance
In computer-integrated manufacturing, machines are equipped with sensors
and memory storage. This provides a large corpus of information retrievable for data
mining and knowledge discovery. The case study is focused on the investigation of
machine breakdown and the effectiveness of scheduled maintenance with the
application of data mining. The research methodology involves eight steps. The initial
step is performed business understanding to gain insight of data mining on machine
performances. Several questions in the stage of macro-level and micro-level are
generated. Second, a proposed simulation model for the operational process was
designed based on a real medical tool manufacturing plant. The production is a job
shop whereby products in batch have to go through a number of processes and multi�stations. Each process alters particular attributes of the product. The production
simulation would be constructed in Witness Horizon V21. Six different machine
breakdown scenarios were modelled. Different feature processing strategies would be
devised, in particular time-related data. Third, a relational database is developed to
store the information from the simulation. The next step is involved data pre�processing which includes data selection, data cleaning and data transformation. Data
mining is the sixth step in which software of Orange is used as the tool. Seventh, the
pattern evaluation is developed to present the discovery of data which helps in
decision-making. From the research, it is found that there have ten types of
breakdowns affecting the performance of machine and the breakdown of coolant
leaking is the main contributor as compared to others breakdown. Besides, the
frequency of breakdown especially for coolant leaking has decreased after the
maintenance is scheduled on the machine. Hence, the application of maintenance on
machine is effective in controlling the frequency of breakdown. From the results of
data, it is able to evaluate the breakdown of machine and support the decision on
scheduling maintenance on machine. However, it requires a large amount of cost to be
invested in maintenance. Last but not least, some of the complex data mining tasks
are not able to perform because of the limited algorithms and machine learning in
Orange software
Spontaneous Subtle Expression Detection and Recognition based on Facial Strain
Optical strain is an extension of optical flow that is capable of quantifying
subtle changes on faces and representing the minute facial motion intensities
at the pixel level. This is computationally essential for the relatively new
field of spontaneous micro-expression, where subtle expressions can be
technically challenging to pinpoint. In this paper, we present a novel method
for detecting and recognizing micro-expressions by utilizing facial optical
strain magnitudes to construct optical strain features and optical strain
weighted features. The two sets of features are then concatenated to form the
resultant feature histogram. Experiments were performed on the CASME II and
SMIC databases. We demonstrate on both databases, the usefulness of optical
strain information and more importantly, that our best approaches are able to
outperform the original baseline results for both detection and recognition
tasks. A comparison of the proposed method with other existing spatio-temporal

Characterisation of protein-truncating and missense variants in PALB2 in 15 768 women from Malaysia and Singapore
Funder: National University Cancer Institute, Singapore; FundRef: http://dx.doi.org/10.13039/501100011105Background: Rare protein-truncating variants (PTVs) in partner and localiser of BRCA2 (PALB2) confer increased risk to breast cancer, but relatively few studies have reported the prevalence in South-East Asian populations. Here, we describe the prevalence of rare variants in PALB2 in a population-based study of 7840 breast cancer cases and 7928 healthy Chinese, Malay and Indian women from Malaysia and Singapore, and describe the functional impact of germline missense variants identified in this population. Methods: Mutation testing was performed on germline DNA (n=15 768) using targeted sequencing panels. The functional impact of missense variants was tested in mouse embryonic stem cell based functional assays. Results: PTVs in PALB2 were found in 0.73% of breast cancer patients and 0.14% of healthy individuals (OR=5.44; 95% CI 2.85 to 10.39, p<0.0001). In contrast, rare missense variants in PALB2 were not associated with increased risk of breast cancer. Whereas PTVs were associated with later stage of presentation and higher-grade tumours, no significant association was observed with missense variants in PALB2. However, two novel rare missense variants (p.L1027R and p.G1043V) produced unstable proteins and resulted in a decrease in homologous recombination-mediated repair of DNA double-strand breaks. Conclusion: Despite genetic and lifestyle differences between Asian and other populations, the population prevalence of PALB2 PTVs and associated relative risk of breast cancer, are similar to those reported in European populations
An outbreak of Streptococcus pyogenes in a mental health facility : advantage of well-timed whole-genome sequencing over emm typing
Financial support: The outbreak investigation was supported by Institute of Mental Health.OBJECTIVE: We report the utility of whole-genome sequencing (WGS) conducted in a clinically relevant time frame (ie, sufficient for guiding management decision), in managing a Streptococcus pyogenes outbreak, and present a comparison of its performance with emm typing. SETTING: A 2,000-bed tertiary-care psychiatric hospital. METHODS: Active surveillance was conducted to identify new cases of S. pyogenes. WGS guided targeted epidemiological investigations, and infection control measures were implemented. Single-nucleotide polymorphism (SNP)-based genome phylogeny, emm typing, and multilocus sequence typing (MLST) were performed. We compared the ability of WGS and emm typing to correctly identify person-to-person transmission and to guide the management of the outbreak. RESULTS: The study included 204 patients and 152 staff. We identified 35 patients and 2 staff members with S. pyogenes. WGS revealed polyclonal S. pyogenes infections with 3 genetically distinct phylogenetic clusters (C1-C3). Cluster C1 isolates were all emm type 4, sequence type 915 and had pairwise SNP differences of 0-5, which suggested recent person-to-person transmissions. Epidemiological investigation revealed that cluster C1 was mediated by dermal colonization and transmission of S. pyogenes in a male residential ward. Clusters C2 and C3 were genomically diverse, with pairwise SNP differences of 21-45 and 26-58, and emm 11 and mostly emm120, respectively. Clusters C2 and C3, which may have been considered person-to-person transmissions by emm typing, were shown by WGS to be unlikely by integrating pairwise SNP differences with epidemiology. CONCLUSIONS: WGS had higher resolution than emm typing in identifying clusters with recent and ongoing person-to-person transmissions, which allowed implementation of targeted intervention to control the outbreak.PostprintPeer reviewe
The Evolution of Primate Short-Term Memory.
Short-term memory is implicated in a range of cognitive abilities and is critical for understanding primate
cognitive evolution. To investigate the effects of phylogeny, ecology and sociality on short-term memory, we tested the largest and most diverse primate sample to date (421 non-human primates across 41 species) in an experimental delayed-response task. Our results confirm previous findings that longer delays decrease memory performance across species and taxa. Our analyses demonstrate a considerable contribution of phylogeny over ecological and social factors on the distribution of short-term memory performance in primates; closely related species had more similar short-term memory abilities. Overall, individuals in the branch of Hominoidea performed better compared to Cercopithecoidea, who in turn performed above Platyrrhini and Strepsirrhini. Interdependencies between phylogeny and socioecology of a given species presented an obstacle to disentangling the effects of each of these factors on the evolution of short-term memory capacity. However, this study offers an important step forward in understanding the interspecies and individual variation in short-term memory ability by providing the first phylogenetic reconstruction of this trait’s evolutionary history. An Indo-Pacifc coral spawning database
The discovery of multi-species synchronous spawning of scleractinian corals on the Great Barrier Reef in the 1980s stimulated an extraordinary effort to document spawning times in other parts of the globe. Unfortunately, most of these data remain unpublished which limits our understanding of regional and global reproductive patterns. The Coral Spawning Database (CSD) collates much of these disparate data into a single place. The CSD includes 6178 observations (3085 of which were unpublished) of the time or day of spawning for over 300 scleractinian species in 61 genera from 101 sites in the Indo-Pacific. The goal of the CSD is to provide open access to coral spawning data to accelerate our understanding of coral reproductive biology and to provide a baseline against which to evaluate any future changes in reproductive phenology
Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)1.
In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide. Our ultimate goal is to encourage intellectual and technical innovation in the field

Non-greedy minimum interference routing algorithm for bandwidth-guaranteed flows
This paper presents a new non-greedy routing and admission control algorithm for the dynamic routing of bandwidth-guaranteed tunnels. The algorithm operates online that is handling requests which arrive one at a time without prior knowledge of the traffic pattern. It combines the key concepts in the minimum interference routing algorithm and the algorithms developed based on theoretic competitive analysis to provide efficient routing. Using extensive simulations, we show that our algorithm out-performs several previously proposed algorithms on several metrics such as the acceptance rate of the tunnel set-up requests, fairness and the capability of providing priorities to tunnel requests of certain source-destination pairs in the network.
